45 CELLULAR IMMUNITY DURING NORMAL PREGNANCY. To investigate a possible role of cellular immunity during normal preg- nancy, monocyte subpopulation,T cell subsets,concanavalin A(Con A) indu- ced suppressor cell activity and natural killer(NK) activity were assessed in peripheral blood of l)normal pregnant women at different stages of gestation and 2)healthy non-pregnant women who were served as control. Monocyte subpopulation and T cell subsets were characterized by indirect immunofluorescence with monoclonal antibody(OKT3,0KT4,0KT8,0KIa l ) and by indirect antiglobulin rossetting reaction(IARR) with OKIa I. Con A induced suppressor cell activity was measured by the suppression of mitogenic proliferation. NK activity was also investigated by~Cr re- lease assay. During the first trimester of pregnancy,a significant increase in the + . . . . percentage of OKT4 T cell and a significant decreas~ in the percentage of OKT8 T cell were observed. The percentage of la monocyte increased significantly during the first and se#ond trimester. The tendency was also observed in the percentage of la- T cell by IARR. Con A induced cell in normal pregnancy suppressed significantly the mitogenic proliferation of both autologous and allogenic peripheral blood lymphocyte. Moreover, NK activity showed a significant decrease during normal pregnancy. Our data suggest that cellular immunity during normal pregnancy may be functionally suppressed via the complicated cell-cell interaction. Nippon Medical School, I-I-5, Sendagi, Bunkyo-Ku, Tokyo, Japan. Aim of study : In the pregnancy C3-system (alternative pathway) of comple- ment will play a important role as primitive defence system. During pregnancy the cellular immunity come down slightly, and the complement system especially C3-system is enhanced in compensation for it. Method : Skin test by DNCB, PPD, and PHA were examined. The complement activity of classical pathway was measured by Mayer's method (CH50) and that of alternative pathway (AI-CH50) was measured by principle that human alter- native pathway hemolysis the rabbit erythrocytes, the component of complement C3,C3-activator, and C4 were measured by single radial immunodiffusion.these measurement were performed in the 82 cases of normal pregnancy, 115 cases of intrauterin fetal death, 28 cases of hydatidiform mole, 21 cases of toxemia, 5 cases of chorionic malignancy and 3 cases of uteroplacental apoplexia. Results : The aged necrotic villi activated the alternative pathway of complement. In the all cases of abnormal pregnancy with necrotic villi, CH50 and AI-CH50 were came down and C3, C3-activator, and C4 were decreased quanti- tatively, especially uteroplacental apoplexia were markedly decreased. Conclusion : In the abnormal pregnancy with low activity of CH50, AI-CH50, and the decrease of C3, C3-activator, and C4, it considered that necrotic villi presented in the uterin cavity and evacuation would become necessary, these findings suggest to resection by nonspecific immune response.
