OBJECTIVES
2:1 randomization
hazard ratio (HR) of 0.625
median PFS1 from 5 months (control arm) to 8 months (olaparib arm)
a total of 193 events (progression or death) in the study would provide a 90% power, a 0.05 1-sided alpha level.
Recruitment duration of 24 months and a 24-month follow-up for the last included patient
297 patients have to be randomized in the study (198 patients in the olaparib arm and 99 patients in the placebo arm) so that maturity of the PFS1 data is approximately 66%.
STATISTICAL PLAN
Overall survival (OS) will be the main secondary
endpoint.
Assuming a 0.625 HR for PFS1 could translate to a
0.70 HR in OS (improvement from 13 months to 18.5
months in terms of median survival), the observation
of 203 deaths among the 297 patients (66% deaths)
will allow to detect such HR of 0.70 with a 73%
power.
