OBJECTIF
Notre équipe a créé un modèle de souris double transgénique conditionnelle (Tet-off/FKBP12.6) dans le fond génétique B6D2. L’expression du transgène est réprimée en présence de doxycycline (Dox) et induite par le retrait de la Dox de l’alimentation des souris. Ces souris devant être soumises à une sténose de l’aorte thoracique (TAC) pour induire hypertrophie et insuffisance cardiaque, nous avons voulu savoir si la Dox exerce un effet sur ces paramètres.
Modèle expérimental: La TAC = surcharge de pression par sténose de l’aorte thoracique réalisée sur des souris mâles B6D2/F1 (19-22g) (n=120) anasthésiées par Kétamine/Xylasine.
Guide d’un diamètre 0,4 mm ( du diamètre ~ 60%).
Cathétérisme du ventricule gauche:Anesthésie: Isoflurane 2%, 100% O2
Introduction de la sonde Millar 1,4 French par la carotide droite jusqu’au ventricule gauche.Logiciel d’analyse : Iox (EMKA).
PCR en temps réel: Extraction et Reverse Transcription des ARNm du ventricule gauche. Les amplifications en temps réel des ADNc du BNP et de la GAPDH sont réalisées dans un Lightcycler (Roche). Le BNP est normalisé par rapport à la GAPDH de chaque échantillon.

Paramètres Hémodynamiques mesurés chez la Souris
� La TAC provoque une hypertension chez les souris t rès marquée à 1 mois et qui diminue à 2 mois.
� Une baisse de contractilité est observée à 2 mois post-TAC, accompagnée d’un dysfonctionnement systolique (Pmax ) et diastoli que (PTDVG ).
� Nous avons défini 2 groupes d’animaux, l’un présen tant une hypertrophie compensée, l’autre une insuffisance cardiaque sévèr e, en se basant sur la présence d’œdème pulmonaire (LW TAC > LWSham + 2 SD).
� Selon ce critère de sélection, un plus grand nombr e de souris est en IC à 1 mois en présence de Dox. Le test de Χ2 révèle que cette différence est significative. Cepe ndant, cette différence disparaît à 2 mois.

Variation des paramètres hémodynamiques chez les souris HC et IC
Une PTDVG très élevée, une dP/dT max et min diminuée sont observées chez les souris en IC,
ce qui reflète un dysfonctionnement cardiaque
sévère et conforte notre classification.
CONCLUSION
La doxycycline favorise le développement de l’hypertrophie et de l’insuffisance cardiaque un mois après une sténose de l’aorte thoracique. Cet effet n’est plus observé deux mois après la sténose.
Ceci doit conduire à la prudence dans la réalisation du plan expérimental et dans l’interprétation des résultats dans les modèles d’hypertrophie cardiaque chez les souris transgéniques dont l’expression du transgène est pilotée par le système Tet-on/Tet-off.
