1. OBJECTIVE
To prepare the smears for staining purpose.

Bone Marrow spreading
3. PRINCIPLE
The smear should have head and trail
4. REQUIREMENTS
4.1 EQUIPMENT
4.1.1 Glass slides
4.1.2 Spreader
4.1.3 Slide holder
4.1.4 Gauze
4.1.5 Petri dish
4.2 SPECIMEN
4.2.1 Aspirated bone marrow4.2.2 Imprint
5 PROCEDURE

5.1 From bone marrow aspiration
5.1.1 Obtain aspirated marrow from the medical personnelimmediately and expel it into petri dish

5.1.2 Rotate the petri dish to look for the marrow fragment MLT/SO
5.1.3 Gently touches the marrow fragment onto a spreader then transferit to the glass slide

5.1.4 Drag the fragment and leave a trail of cells behind the spreader MLT/SO
5.1.5 Repeat the step 5.1.3 to 5.1.4 to get at least 7 slides for newcases and at least 3 slides for old cases MLT/SO
5.1.6 Label the slides with
i) Patient R/N and
ii) Date of sample collection

5.1.7 Allow the slides to dry in the air before stain MLT/SO
5.2 From imprint
5.2.1 Obtain bone biopsy from the medical personnel and placed it inthe middle of one glass slide,
MLT/SO5.2.2 Placed a second slide on top of the first slides and rolled it
forward and backward gently for few times
MLT/SO5.2.3 Repeated step 5.2.2 for few time to get at least 2 slides MLT/SO5.2.4 Label the slides with
iii) Patient R/N and
iv) Date of sample collection

5.2.5 Allow the slides to dry in the air before stain MLT/SO
5.3 For storage5.3.1 Wrap tightly the additional air-dried unfixed and unstained
smear with in aluminum foil
MLT/SO5.3.2 Label it with
i) Patients name
ii) Patients R/N
iii) Date of sample collection
MLT/SO
5.3.3 Storage it at 4oC-10
oC for future use purposes MLT/SO
