植物の系では外部環境への応答や適応、光合成、植物
ホルモンによる成長制御、高効率での分化全能性など動
物の系とは明らかに異なった植物独自の個体レベル、細
胞レベル、分子レベルでの制御機構が存在している。
当協力研究グループではモデル植物のシロイヌナズナ
を用いて遺伝子破壊型変異体や過剰発現型変異体(ミ
ニ FOXシステム)のリソース開発、シロイヌナズナ・
ポプラ・ダイズ・コムギ・その他植物種の完全長 cDNA

ス業務に貢献している。さらに、表現型、遺伝子発現、
代謝産物、タンパク質などをそれぞれ網羅的に解析する
「オミクス解析」および、個々の遺伝子の詳細な解析に
よって得られた情報を統合することで、これらリソース
に付加価値をつける。また、環境ストレス応答機構の研
究を通じて人類が直面している環境・食料問題に取組ん
でいる。

年次計画と成果
1. トランスポゾン(Ds)挿入シロイヌナズナ変異体コレクションの表現型解析(フェノーム解析)
ポストゲノム解析として我々は、シロイヌナズナ遺伝
子へのトランスポゾン Ds挿入変異体作製に引き続き、
Dsの同型接合体(ホモライン)を用いてフェノーム解
析(全遺伝子破壊変異体を用いた網羅的表現型解析)を
行っている。形態変化を指標にした表現型解析について、
我々が作製した約 18,000 Ds挿入変異体コレクションの
うち約 12,000ラインの解析は前年度に報告した。今年
度からは残り 6,000ラインの解析を開始し、現在継続中
である。
フェノーム解析の過程で、雄性不稔を示す male
sterility1 (ms1)変異体(図 1)を単離し、今年度はその
詳細な機能解析を行った。雄性不稔の原因遺伝子 MS1
は転写活性化因子をコードし,花粉成熟に必要な下流遺
伝子を制御していることが明らかとなった。

ムシグナル伝達とのクロストークについて研究を行っ
た。また、ストレス耐性獲得のシステムを明らかにする
ために、ストレス応答性カルシウム結合タンパク質の機
能解析、ストレス応答性アブシジン酸(ABA)合成酵
素の発現制御解析、ユビキチン化を介したストレス応答
性転写因子の分解系について研究を行った。
5. ストレス耐性植物の実用化に向けた形質転換作物の作製と圃場試験
本研究グループではこれまでに、環境ストレス耐性に
関与する遺伝子を多く単離してきた。本研究では、これ
ら耐性遺伝子を利用したストレス耐性作物の実用化を目
指し、主要作物の一つであるコムギに注目して研究を
行っている。本年度は、コムギに乾燥耐性を付与させる
ための最適な遺伝子をシロイヌナズナより選抜し、導入
用ベクターを作製した。また乾燥ストレス付与に関わる
38遺伝子の完全長 cDNAを、コムギ相同遺伝子として
収集した。更にコムギのマイクロアレイ解析情報などを
用いて、恒常的発現型プロモーターの選抜を行った。今
後はこれらを組み合わせてコムギ品種等に導入し、得ら
れた形質転換体について圃場条件における乾燥耐性や収
量等を評価していく(図 3)。
6. トランスクリプトミクスとメタボロミクスの統合による ABA依存的な乾燥ストレス応答性代謝ネットワークの解明
乾燥ストレス下の植物において、ABAは細胞内シグ
ナル伝達や多くの遺伝子の発現誘導に重要な働きを行
う植物ホルモンである。本研究では、シロイヌナズナ
ABA合成関連遺伝子 NCED3の破壊株を実験材料とし、
トランスクリプトミクスとメタボロミクス解析を組み合
わせることで、乾燥ストレス下で合成された ABAが制
御する代謝ネットワークの包括的な解明を行った。
2. ダイズ完全長 cDNAプロジェクト
理研 PSCでは、ポストゲノム研究にとって有用なリ
ソースとなる完全長 cDNAの整備を、複数の有用作物
について進めている。本研究グループでは、代表的なマ
メ科作物であるダイズの完全長 cDNAプロジェクトを
担当し、約 40,000クローンについて塩基配列の収集お
よびデータ解析を終了した。現在、全てのデータの公開
に向けたシステムを構築中である(図 2)。
3. 過剰発現型トランスジェニック植物を利用したミニ FOXシステムの開発
本研究グループでは、次世代型アクチベーションタ
ギング法である FOX Hunting System(full-length cDNA
over-expressor gene hunting system)を利用して、環境ス
トレス応答性を示すシロイヌナズナ転写因子の機能解析
を行った。41個の環境ストレス誘導性転写因子の完全
長 cDNAを高発現する植物ライブラリーを作製し、塩
ストレス耐性に関与する bZIP60遺伝子の単離に成功し
た。
4. 環境ストレス応答に関わる遺伝子の機能解析 植物は乾燥、塩、低温ストレスなどの様々な環境スト
レスに応答して、植物ホルモンを介したシグナル伝達と
遺伝子発現を行い環境に適応している。本研究グループ
では植物の生産力向上のため、環境ストレス応答に関連
した遺伝子探索と機能解析を中心に研究を行っている。
本年度は環境ストレス応答におけるシグナル伝達経路を
明らかにするために、環境ストレスと植物ホルモンで活
性化するプロテインキナーゼの機能解析および、ジャス
モン酸シグナル伝達に関わるMAPキナーゼとカルシウ
