Abstract

ThehumanGATA1,hGATA1(previouslycalledNFE1,GF1orEryf1),amajorsequencespecificDNAbindingproteinoftheerythrocyticlineage,isamemberofazincfingerfamilyofDNAbindingproteins.WereportherethecloningofahumancDNAforanewmemberofthisfamily.Thismember,calledhGATA3,has85%aminoacidhomologywithhGATA1intheDNAbindingdomainandnohomologyelsewhereintheprotein.UnlikehGATA1,hGATA3isnotlocalizedontheXchromosomeandwemapittothe10p15bandofthehumangenome.NorthernblotanalysisindicatesthatthisfactorisaTcellspecifictranscriptionfactor,presentbeforeactivationandupregulatedduringTcellactivation.TheencodedhGATA3protein,madeinaninvitrotranscriptiontranslationassay,bindstheWGATARmotifpresentinthehumanTcellreceptor(TCR)deltageneenhancerand,bytransfectioninHeLacells,weshowthathGATA3cantransactivatethisTCRdeltageneenhancer.InterestinglythisenhancerbindsandisalsotransactivatedbyhGATA1.Conversely,thepromoterofthehumanglycophorinB(GPB),whichiserythroidspecificandcontainstwoWGATARmotifs,bindsandistransactivatedbyhGATA1and,toalesserextent,byhGATA3.TheseresultsindicatethattheactivationofspecificgenesbyhGATA1orhGATA3ispartlygovernedbythelineageexpressionofthesetwofactorsduringhaematopoiesisandthat,intheTcelllineage,hGATA3bindsthehumanTCRdeltageneenhancerandisinvolvedinitsexpression.
