Non-leaky Expression
• Ein Expressionsytem soll möglichst “dicht”- non-leaky sein.
• Grund: Proteine, die exprimiert werden sollen, können toxisch sein.
• Das kann zur Folge haben, dass sich der Expressionshost gar nicht transformieren lässt, da die transformierten Zellen sofort sterben.
• Stirbt der Expressionshost nicht, kann aber das Zeilprotein, das durch leaky Expression produziert wird, eine negative Selektion gut exprimierender Zellen bewirken. Erklärung: Gut exprimierende Zellen produzieren viel Zielprotein, aber weniger eigene Proteine, d.h. wachsen schlechter. Die gut exprimierendensind nach einigen Verdopplungsrunden fast nicht mehr vorhanden. Man selektioniert also auf schlecht exprimierende Zellen.
• Das pET System von Studier für Expression in E.coli ist extrem “dicht” aufgrund verschiedener Vorkehrungen.

Keine Expression ohne Induktion
• da lac-Repressor Protein auf Operatorregion sitzt und Bindung der E.coli Polymeraseverhindert, d.h es wird keine T7 Polymerasesynthetisiert.
• Ohne T7 Polymerase keine Expression des Zielproteins.
• Als zusätzliche Sicherung sitzt auch beim T7 Promoter noch eine lac Operatorregion. Sollten T7 Polymerase dennoch in geringen Mengen synthetisiert worden sein, kann das Gen trotzdem nicht abgelsen werden.
• Zusätzliche Kopien des Lac Repressors sind Plamid kodiert. So wird genügend lac Repressorsynthetisiert.

Zusätzliche Sicherung
• Eine weitere Absicherung gegen T7 Polymerase, die in geringen Mengen synthetisiert wurde, ist die Co-Expression von Lysozym, welches auch als Inhibitor der T7 Polymerase wirkt. Die T7-Polymerase wird abgefangen, bevor die Expression des Zielgensstattfindet. • Das Lysozym wird auf einem zweiten Vektor kodiert.

-Use protease inhibitors
-Work fast (prepare buffers, SDS –PAGE, Blots etc)
-Keep cold protein (slower denaturation and less proteolysis)
-Do NOT freeze in between; if necessary shock freeze with glycerol.

-Make small scale tests prior large scale preparations
