PECULIARITY OF THE CIACCIO-POSITIVE

LIPID IN THE CAT ADRENAL CORTEX

The mammalian adrenal cortex, despite excep­tions, is histochemically a lipid rich structure. And it is assumed that fluctuations in the intensity of lipid staining of the adrenal cortex indicate the variety of the cortical activity; lipid depletion denotes hyperactivity and lipid accumulation hypo­activity of the gland.

In contrast, some ruminants and hamsters have a different kind of adrenal cortex. The gland of this kind is negative to the SCHULTZ test, and con­tains few, if any, lipid droplet. This "poor lipid droplet" is normally observed only in the zona glomerulosa and increases in the same zone under stressed conditions.

The present author has found that the distribu­tion of the lipid droplet indicated by the CIACCIO method in the lipid rich adrenal of the dog is similar to that of the poor lipid observed in the lipid poor adrenal as the hamster gland.

In this experiment, cat adrenals were used in order to study the alteration of the CIACCIo-positive lipid. The cat qualifies as having lipid rich adrenals.

Methods

The animals used were six weaned cats. All animals were divided into the following three in­stances to inspect the alteration (If the cortical lipid. Two animals served as controls. And two animals were stressed; one animal drowned until just before the death and the other was deprived of all nutrient for seven days. The rest animals were given daily subcutaneous injections of 0.5mg. cortisone acetate for seven days.

All animals were sacrificed by a stab into the heart. The adrenals removed, trimmed free of all superfluous tissue, weighed on a torsion balance and were fixed promptly. The right adrenals were placed in 10% formalin. And after several days fixation, frozen sections were cut at 5", and staind with Sudan black B, Nile blue SUlphate and by the method of SCHULTZ. The left adrenals were treated by the CIACCIO procedure; the staining was accom­plished with Sudan black.

Observations

The unstimulated cat gland contains many sudano­philic lipid droplets, which are irregular in the shape. And the intensity of the Sudan stain parallels with the intensity of the SCHULTZ reac-

tion. These observations denote the cat adrenal to be lipid rich type in both different groups of mammalian adrenal cortex.

In the adrenal of a drowned cat, there was a evident decrease in sudanophilic lipid in the zona fasciculata. In the zona glomerulosa the lipid alter­ation is not marked. The SCHULTZ test is most reduced in the outer fasciculata.

Acute starvation is accompanied by low amount of sudanophilic lipid in the zona fasciculata. But the lipid reduction is slight. The SCHULTZ reaction is slightly present in the same zone.

Repeated injections of cortisone acetate cause adrenal atrophy. After the injections for seven days the sudanophilic lipid is depleted considerably all over the cortex. The SCHULTZ reaction occurs in all zones.

In general, the amount of lipid shown by the CIACCIO method is extremely less than the total lipid content as determined by the Sudan stain, since the greater part of sudanophilia in the corti­cal cells is soluble in the process of paraffin imbedding.

In the normal gland, it is next to impossible to find the CIACCIO-positive lipid droplets, though the connective tissue and protoplasm are colored lightly. In the drowned cat adrenal the CIACCIO-positive lipid droplets appear in large numbers in the zona fasciclliata which exhibits the reduction of sudano­philia in frozen section. These CIACCIO-positive lipid take the form of ring or spheroid and their diameters are ranged from 3", to the limit of resolution of the light microscope. In addition to these observations. they are stained rose color with Nile blue sulphate. The accumUlation of this lipid in the zona fasciculata is observed also in the acutely starved cat, however the form of the lipid droplet is irregular.

In the gland of the cortisone treated cat the CIACCIO-positive lipid droplet is scarecely recog­nized.

Discussion

In the mammalian adrenal cortex, lipid droplet as indicated by the method of CIACCIO is scarecely investigated, though sudanophilia in the frozen section is interpreted by many workers. For that reason the alteration of the CIACCIO-positive lipid is most interesting, while the alteration of the

sudanophilia needs not discuss. In recent years, KNIGGE!) and DEANE and LYMAN2)

pointed out that the alteration of the "poor lipid" proved in the hamster adrenal cortex is different from that of "rich lipid" as observed in the rat adrenal. The present author has described that the goat adrenal is histochemically similar to that of the hamster and that the poor lipid in the goat cortex is represented by the method of CIACCIO~·4) By the latest work of the same author. it is found that the distribution of the CIACCiO-positive lipid sorted out from rich lipid in the dog adrenal dif­ferent from that of the sudanophilia in the frozen section and shows the same alteration as the poor lipid pointed out by the above workers. It is probably safe to assume that the lipid droplet indicated by the method of CIACCIO is the most indispensable lipid for the production of cortical hormone since this lipid alone is common in both types of lipid rich and lipid poor adrenal.

Now, in the cat adrenal used in this experiment the distribution of the sudanophilia observed in the frozen section is all similar to those of the dog and rat. However, the distribution of the lipid produced by the method of CIACCIO differs from those of the dog and rat. This lipid in the cat adrenal is accumulated under stress in the zona fasciculata and does not appear after Cortisone treatment. This fact exhibits that all mamalian adrenal corticies do not make the arrangement of the CIACCIO-positive lipid uniform. Neverthress, it is most striking to note that the CIACCIO-positive

lipid increases under stress in all animals experi­mented. The singnificance of the existence of the CIACCIO-positive lipid must be revealed in furture.

Summary

A study on the alterations of the CIACCIO-positive lipid under stress and cortisone treatment was made in the adrenals of the cat a few months of age.

The distribution of the CIACCiO-positive lipid is quite different from that of the sudanophilia ob­served in frozen section. In the cat the lipid droplet produced by the method of CIACCIO is in­creased in the zona fasciculata under stress and disappears by the treatment of cortisone acetate.

Such alteration of this lipid observed in the cat does not come up to that of the same lipid in the dog investigated previously by the present author.

If it be so, I t is concluded that the distribution of the CIACCiO-positive lipid in adrenals differs pro­bably according to mammalian species.
