Update on BSD® Punchers by Luminex

• On April 28, 2014 Luminex communicated to Life Technologies the plan to cease the operations of BSD and the manufacture of BSD® punchers

• On the same date, Luminex announced they would no longer accept any BSD® Puncher orders

5

Life Technologies and BSD® Punchers

• Life Technologies has been a distributor of BSD® Punchers in the forensic market since the end of 2009

• Life Technologies recognizes the importance of service and support for deployed BSD® instruments

• Life Technologies plans to continue to offer its standard comprehensive service plan for the BSD® punchers until June 30th, 2016

• As a result of the Luminex decision, Life Technologies may need to offer more limited service

Life Technologies & Copan

• Life Technologies started distributing Copan sample collection devices for forensic applications including the NUCLEIC-CARD™ system in 2012

• Life Technologies and Copan signed a distribution agreement in June 2014 to distribute the CPA technology developed by Copan

• Life Technologies is now the global exclusive distributor of CPA instruments in the human identification market

• Life Technologies will provide service and support for installed CPA instruments

CPA200 – Supported Cards

Optimized for NUCLEIC-CARD™ with Cleaning Area

• Validated punch size: 1.2 mm

• Additional punch sizes are currently under consideration

• Protocol with “cleaning punch” available

CPA200 works with sample collection systems routinely used in forensic laboratories: • NUCLEIC-CARD™ (regular and custom) • FTA® Cards (Whatman) • Buccal DNA Collector (Bode)

10

CPA200 – Punch Quality

CPA200 includes: • Ionizer – to neutralize the electrostatic charges on the

puncher tool to ensure detachment of paper disks and release in the correct well

• Dust Remover – to remove any card debris from the puncher tool to ensure excellent cleaning conditions and optimal cutting quality

CPA200 – Cross-Contamination Study

• Objective: Assess well cross-contamination when preparing a 96-well plate with CPA200

• Method: Collect samples from 23 donors using FLOQSwabs™ Regular Transfer samples onto 1 spot NUCLEIC-CARD™ COLOR, 4 spots

NUCLEIC-CARD™ COLOR, FTA® cards and Bokumbio cards. Cards were dried at RT for one hour Add 15 µL GlobalFiler® Express Reaction Mix1 to each well of the 96-

well plate Punch one 1.2 mm disc in each well of the plate followed by one

cleaning strike between punches Seal the plate and centrifuge it at 300 rpm for 20 seconds Amplify the samples using a GeneAmp® PCR System 9700

15

CPA200 – Key Findings

• Good punch quality with different card types • No instances of incorrect barcode reading • No disc losses • Consistent release of the paper discs at the bottom of the

well as confirmed by the real-time feedback from the camera • No instances of cross-contamination • Full concordance between STR profile and expected profile

from the donor1 • Very user-friendly software

1. Samples from each donor were spotted on two cards. Each card was punched so that the discs associated with each donor randomly ended up in two different plate wells

CPA300 – Punching Unit

• Puncher has two main functions: generates a 1.2 mm disk that

is safely released in the 96 well plate performs the cleaning

procedure. The cleaning disk(s) are released in a separate bin

• Ionizer removes the electrostatic

charges

• Dust Remover removes card debris

Two cards per minute WITH

cleaning punch

Page 21: Introducing CPA200 and CPA300 Punching Systems

22

CPA300 – Plate and Card Grippers

• Plate gripper enables the movement of the plate during the punching and liquid handling procedures

CPA300 can accommodate up to three 96-well plates

Bin to collect cleaning punches

• Card gripper transports the card from and to the card rack to enable the punching procedure

CPA300 can accommodate up to four card racks1

1. Each rack fits up to 75 cards

Page 22: Introducing CPA200 and CPA300 Punching Systems

26

Sealing Unit

• Plates are automatically sealed at the end of the liquid handling procedure

Reduces risks of cross-contamination Helps prevent reagent

evaporation PCR-ready plate for better

efficiency

Full cycle: Approx. one hour per plate1

1. Includes punching, cleaning, liquid handling and sealing
