Le basi biologiche delle patologie croniche e degenerative dell'adulto e dei
disturbi neurologici dell'infanzia: qual è il nesso comune?
Un importante dilemma della ricerca scientifica attuale è se esistano “poche malattie”
ma molte tipologie di individui oppure “molte malattie” ma poche tipologie di
individui.
Lo sguardo “oggettivante” della medicina occidentale ha favorito la “teoria delle
molte malattie”, ha cioè determinato una parcellizzazione dei disturbi e della malattie
ed un ultra-settorializzazione delle competenze mediche a scapito della visione
d'insieme dell'individuo come organismo inserito in dimensione ecologica.
La relazione verterà d'altro canto sullo scandagliare l'ipotesi delle “poche malattie”
in organismi con caratteristiche differenti. Saranno descritte le recenti ricerche
scientifiche che evidenziano le connessioni causali che accomunano gran parte delle
patologie croniche tumorali, degenerative ed infiammatorie. Infine si cercherà di
indicare quali possono essere le caratteristiche metaboliche comuni che inducono in
individui diversi manifestazioni patologiche differenti e quali le ipotesi di trattamento
biomedico efficaci.

Un importante dilemma della ricerca scientifica attuale è se esistano “poche malattie” ma molte tipologie di individui oppure “molte malattie” ma poche tipologie di individui.

Risultati: Osservando i bambini all'età di 8 anni, Circa 1% dei bambini ossia 1 su 110 bambini in media negli USA risulta avere una diagnosi di Disturbo dello Spettro Autistico.In rapporto al 2002 il tasso di incremento è del 57% . Sebbene ci possa essere un miglioramento della capacità diagnostiche, non può essere negato che ci sià un reale aumento del rischio di sviluppare ASD.Questi risultati indicano una aumento della prevalenza di Autismo negli USA e sottolineano la necessità di considerare l'Autismo come un urgente problema di salute pubblica!È inoltre necessario un monitoraggio continuo per documentare quali siano i fattori di rischio che possano contrubuire allìinsorgere dell'Autismo

Glutamate neurotoxicity has been implicated in stroke, head trauma, multiple sclerosis and neurodegenerative diseases. Although recent data show that cultured glioma cells secrete glutamate, the growth potential of brain tumors has not yet been linked to an excitotoxic mechanism. Using bioluminescence detection of glutamate release from freshly prepared brain slices, we show that implanted glioma cells continue to secrete glutamate. Moreover, gliomas with high glutamate release have a distinct growth advantage in host brain that is not present in vitro. Treatment with the NMDA receptor antagonists MK801 or memantine slowed the growth of glutamate-secreting tumors in situ, suggesting that activation of NMDA receptors facilitates tumor expansion. These findings support a new approach for therapy of brain tumors, based upon antagonizing glutamate secretion or its target receptors.

La neurotossicità del glutammato è stato riconosciuto nell'infarto, trauma cerebrale, sclerosi multipla e nelle patologie neuro-degenerative. Ma anche la crescita del Glioma Maligno è favorito nella sua crescita dalla presenza di glutammato. L'utilizzo di farmaci che bloccano i recettori del glutammato riduce la progressione di crescita del tumore.

The concept that bioactive components in functional foods are efficacious for the improvement of health, has recently gained much importance. The cruciferous vegetables which include broccoli, cabbage and
cauliflower are excellent source of phytochemicals including glucosinolates and their byproducts, phenolics and antioxidant vitamins as well as dietary minerals. Broccoli consumption mediates a variety of functions
including providing antioxidants, regulating enzymes and controlling apoptosis and cell cycle. The organosulfur chemicals namely glucosinolates and the S-methyl cysteine sulphoxide found in broccoli in
concert with other constituents such as vitamins E, C, K and the minerals such as iron, zinc, selenium and the polyphenols namely kaempferol, quercetin glucosides and isorhamnetin are presumably responsible for various health benefits of broccoli. There exists no comprehensive review on the health promoting effects
of phytochemical compounds present in broccoli so far. This review compiles the evidence for the beneficial role of glucosinolates in conjugation with the other phytoconstituents for human health. It also gives an overview on the chemical and biological characterization of potential bioactive compounds of broccoli
including the interaction of phytoconstituents on its bioactivity. Further, the molecular basis of the biological activities of the chemicals present in broccoli potentially responsible for health promotion, from
chemoprevention to cardio protection, are outlined based on in vitro and in vivo studies with a note on the structure activity relationship of sulforaphane and a few other isothiocyanates.

Oxidative stress is an important molecular mechanism of astrocyte injury and death following ischemia/reperfusion and may be an effective target of intervention. One therapeutic strategy for
detoxifying the many different reactive oxygen and nitrogen species that are produced under these conditions is induction of the Phase II gene response by the use of chemicals or conditions that promote
the translocation of the transcriptional activating factor NRF2 from the cytosol to the nucleus, where it binds to genomic antioxidant response elements. This study tested the hypothesis that pre- or post-treatment of cultured cortical astrocytes with sulforaphane, an alkylating agent known to activate the
NRF2 pathway of gene expression protects against death of astrocytes caused by transient exposure to O(2) and glucose deprivation (OGD). Rat cortical astrocytes were exposed to 5 muM sulforaphane
either 48 h prior to, or for 48 h after a 4-h period of OGD. Both pre- and post-treatments significantly reduced cell death at 48 h after OGD. Immunostaining for 8-hydroxy-2-deoxyguanosine, a marker of
DNA/RNA oxidation, was reduced at 4 h reoxygenation with sulforaphane pretreatment. Sulforaphane exposure was followed by an increase in cellular and nuclear NRF2 immunoreactivity. Moreover, sulforaphane also increased the mRNA, protein level, and enzyme activity of NAD(P)H/Quinone
Oxidoreductase1, a known target of NRF2 transcriptional activation. We conclude that sulforaphane stimulates the NRF2 pathway of antioxidant gene expression in astrocytes and protects them from cell
