コクサッキーウイルスとアデノウイルスの共通受容体 CAR について

Abstract
The pituitary is a major endocrine organ and produces many hormones important in response to various phys-
iological processes. The maintenance of postnatal pituitary depends on the continuous supply of functional cells
by stem/progenitor cells throughout life. In recent years, the research on a stem/progenitor cell niche in the pit-
uitary has advanced remakably. We recently found that Coxsackievirus and Adenovirus Receptor (CAR) plays
a role in maintaining the pituitary stem cell niches. In this paper, we review the studies of CAR on its molecular
structures and physiological roles.
要 旨 下垂体は,多くの生体機能を制御する各種ホルモンを合成・分泌する主要な内分泌器官である。下垂
体組織の恒常性維持は,幹・前駆細胞が最終分化細胞であるホルモン産生細胞を絶えず供給することで保たれ
ている。最近,この下垂体の幹・前駆細胞が存在する場所であるニッチに関する研究が進んでいる。そのよう
な背景の中,我々はコクサッキーウイルス・アデノウイルス受容体(Coxsackievirus and Adenovirus Recep-
tor, CAR)が下垂体ニッチにおいて,幹・前駆細胞の接着・移動や幹細胞性の維持を制御する重要な役割を担
っている可能性があることを報告した。本稿では,CAR の分子構造の特徴や生体内における生理機能に関す
るこれまでの研究を概説する。
. はじめに
下垂体は,主要な内分泌器官の一つであり,全ての
脊椎動物の脳の直下に共通して存在している。この組
織は,上皮組織由来の腺下垂体を構成する前葉・中葉
と,神経組織由来の後葉の,3 葉から構成されてお
り,成長,生殖,適応,代謝,行動,ストレス,浸透
圧など多くの生体機能を制御する複数のホルモンを合
成している(Zhu et al. 2005加藤 et al. 2009諏佐
et al. 2007)。
最近,筆者らは,CAR がラット下垂体の幹・前駆
細胞に存在して幹細胞ニッチである Marginal Cell
Layer (MCL)を形成し,さらに,生後直後に MCL
における CAR 陽性細胞は上皮間葉形質転移により
実質層へ移動し,新たな実質層ニッチを形成し,成熟
下垂体の組織形成と構成細胞の供給に機能しているこ
とを論文報告した(Chen et al. 2013)。また,CAR の
発現は胎仔期の様々な組織で高く,出生後には低くな
ることや,CAR のノックアウトマウスは胎性致死を
示すことが知られており,組織形成に深く関わってい

ることが予想される。そこで本稿では,CAR の発見
に至るまでの経緯,分子構造,そして機能について,
これまでの研究をとりまとめて概説する。
. CAR の発見
ウイルスは,細胞膜に存在する受容体を介して感染
する。1976年,LonbergHolm らが,異なるウイル
スファミリーに属するコクサッキーウイルス B 型
と,アデノウイルスの 2 型と 5 型が,共通の膜レセ
プターに結合することを報告した(LonbergHolm et
al. 1976)ことから,CAR に関する研究が始まった。
1985年になって,Mapoles らは,HeLa 細胞(ヒト子
宮頸がん細胞株)からコクサッキーウイルス B3 型と
結合する約 49.5 kDa の膜受容体タンパク質を初めて
単離し,Rpa と命名した(Mapoles et al. 1985)。さ
らに,12年の時を経た1997年に,Bergelson らは,免
疫アフィニティークロマトグラフィーを用いて,
HeLa 細胞からコクサッキーウイルス B 型の膜受容体
タンパク質を精製し,その受容体のアミノ酸配列を明
らかにした(Bergelson et al. 1997)。その膜受容体タ
ンパク質をコードする遺伝子が同定され,ヒトゲノム
中に同定されていたアデノウイルス受容体と一致する
ことが判明した(Mayr & Freimuth 1997)。コクサッ
キーウイルスとアデノウイルスはカプシドの外側に
あるファイバータンパク質を介して,宿主細胞に存
在する受容体 CAR と結合することにより,細胞表面
に吸着して細胞内部に侵入する(図 1)(Khurana &
Meyer 2003)。こうした経緯から,この膜受容体は,
コクサッキーウイルスとアデノウイルスの共通受容体
として,Coxsackievirus and Adenovirus Receptor
(CAR,遺伝子名は Cxadr)と命名され,その後,多
くの研究者によって,その生物学的機能の解析が進め
られてきた(Bergelson et al. 1997Bowles et al.
1999Hauwel et al. 2005Khurana & Meyer 2003)。
. CAR の分子構造
CAR 遺伝子(Cxadr)は,8 個のエクソンから構成
されている。CAR には,選択的スプライシングによ
るアイソフォームの存在が報告されている(Chen et
al. 2003)。マウスでは,2 種のアイソフォーム,
CAR1 と CAR2 が存在し,mRNA の長さはそれぞれ
1.9 kb と 5.7 kb であり,いずれも膜外ドメイン,膜
貫通ドメイン,膜内ドメインを含んでいる(図 2A)。
その二つアイソフォームでは,エクソン 16 がコー
ドする膜外と膜貫通ドメインは同じであるが,エク
ソン 7 と 8 がコードする膜内ドメインの配列の一部
が異なっている(Chen et al. 2003)。CAR のアミノ酸
配列は,N 末端側から,短いリーダー配列(SS,19
残基),膜外ドメイン(D1 と D2,216残基),膜貫通
ドメイン(TM,23残基),および,膜内ドメインの
カルボキシル領域(94あるいは107残基)で構成され
ている(図 2A)。ヒトの場合,膜貫通型 CAR1 と
CAR2 以外に,選択的スプライシングにより膜貫通領
域を欠いた可溶型分子も報告されている(Thoelen et
al. 2001)。
CAR は免疫グロブリンスーパーファミリー(IgSF)
に属する分子であり,タンパク質の高次構造の解析に
より,N 末端側の膜外ドメインには,2 つの Ig 様構
造を持っている事が判っている。それぞれ遠位の D1
(Vlike module)と近位の D2 (C2like module)と
呼ばれており,他分子と相互作用する領域である
(Coyne & Bergelson 2005)。CAR を含むこうした類
似構造を持っているタンパク質は,最初にアフリカツ
メガエル(Xenopus laevis)の皮質胸腺細胞(cortical
thymocytes)から見つかった分子 CTX (Cortical
Thymocyte Marker in Xenopus)から命名された
CTX ファミリーに分類された(Chretien et al. 1996
Du Pasquier et al. 1999Raschperger et al. 2004)
(図 2B)。CTX とは,Ig ドメインを持つ IgSF の原型
と考えられている(Chretien et al. 1998)。生体内に
おける CAR は,この領域を介して,自己会合
(homophilic binding)や,他のタンパク質との相互作
用(heterophilic binding)により複合体を形成するこ
とが確認されている(Patzke et al. 2010)。CAR 同士
の自己会合では,同一細胞内の分子は膜外ドメイン頭
部の D1 同士,あるいは,尾部の D2 同士が向かい合
って結合する形式により,U 形のホモ二量体形成を
行う(図 2C)。隣接する細胞の CAR 分子は,主とし

A. CAR 遺伝子は 8 個のエクソンから構成されている。アイソフォーム CAR1 と CAR2 は,エクソン 16 がコードする膜外と膜
貫通ドメイン配列は同じであるが,エクソン 7, 8 がコードする膜内ドメインの配列が異なる。CAR のアミノ酸配列は,N 末端
側から,膜外ドメイン(短いリーダー配列(SS)と D1, D2 からなる),膜貫通ドメイン(TM),および,膜内ドメインなどの
特徴的な構造で構成されている。文献(Tomko et al. 1997)より許諾を得て掲載。B. CTX 膜タンパク質は,免疫グロブリンスー

A. アイソフォーム CAR1 と CAR2 の C 末端は,異なる PDZ 結合ドメインを持つ。CAR1 は ITVVCOOH を,CAR2 は GSIV

て,D1D2 の相互作用によりタイトジャンクション
を形成する(Patzke et al. 2010)(図 2C)。一方,
CAR 膜外ドメインは,他の分子との異種間結合を形
成することで,多様な機能に関連している。細胞外マ
トリックスとの結合では,CAR の膜外ドメイン D1
D2 を介して,マトリックス糖タンパク質 TNR,
LN1, agrin などと異種間結合を形成している。こう
した結合が,細胞接着や神経突起のガイダンスに関わ
るとの報告がある(Patzke et al. 2010)。また,CAR
の異種間結合形式がウイルス付着に関与しており,主
に CAR の D1 ドメインがアデノウイルス殻外結合部
分とヘテロダイマーを形成することが示されている
(Bewley et al. 1999van Raaij et al. 2000)(図 2D)。
CAR の膜内領域には,PDZ ドメインに結合するド
メイン(PDZ 結合ドメイン)が存在する。PDZ ドメ
インとは,およそ90アミノ酸残基からなり,Pos-
tsynaptic density 95 (PSD95), Drosophila disc large
tumor suppressor (Dlg1), Zonula occludens1 (ZO1)
に共通して存在することから名付けられ(Kennedy
1995),多くのタンパク質に存在する結合モジュール
である。その中には,多様なシグナル伝達に関わるタ
ンパク質が含まれ,その PDZ ドメインを介して,細
胞内における PDZ 結合ドメインを持つタンパク質と
相互作用することにより,細胞内シグナル伝達など,
重要な機能を担っていると考えられている(Ran-
ganathan & Ross 1997)。確認された CAR のアイソ
フォームの C 末端は,いずれも PDZ 結合ドメインを
持つが,アミノ酸配列は異なる。CAR1 は ITVV
COOH を,CAR2 は GSIVCOOH の配列を持ってい
て(Raschperger et al. 2006)(図 3A),この違いによ
り相互作用する細胞内 PDZ ドメインタンパク質に差
異が生じ,それぞれが異なる生理機能に繋がっている
可能性が示唆されている。後述するように,このアイ
ソフォームの差異が CAR の細胞内局在に関連してい
ることは大変興味深い(ExcoŠon et al. 2010)(図 3B)。
. CAR の発現組織と機能
CAR は多様な生物種でその存在が確認されてい
る。哺乳類,鳥類,爬虫類,両生類,条鰭亜綱等の種
を含めてアミノ酸配列の高い保存性を示し,特に,哺
乳類であるヒト,チンパンジー,ウシ,イヌ,マウス
とラットの種間では,90以上の相同性が保持され
ている(Bergelson et al. 1998Fechner et al. 1999
Tomko et al. 1997)。こうした進化上の高い保存性は,
CAR が重要な役割を担っている事を示すもので,こ
れまでいろいろな生理機能が報告されている(Coyne
& Bergelson 2005)。
CAR は,発生中の神経管の上皮細胞で強い発現を
示し,細胞間の相互作用,分裂,移動,さらに,神経
線維束の形成などに関与することが報告されている
(Honda et al. 2000)。心臓においても,CAR の高い
発現が胎仔期から出生後初期まで観察され,細胞間接
着を介して心臓の組織あるいは心筋細胞の形成と修復
に関わっていると考えられている(Kashimura et al.
2004Fechner et al. 2003)。また,CAR は精巣の形
成初期から生殖細胞とセルトリ細胞で強く発現し,精
巣上皮のタイトジャンクションを構成して血液精巣関
門を介する精母細胞の通過に関与していることが示さ
れている(Mirza et al. 2005Meng et al. 2005)。そ
の他に,胎仔期から広範な組織で強く発現している事
も観察されている(Hotta et al. 2003)。
遺伝子改変マウスは生物機能を探索するための重要
な方法であり,CAR の欠損マウス(CAR-/-)が作
製されている。しかし,このマウスは発生初期
(E9.5E13.5)で胎性致死を示した(Chen et al. 2006
Dorner et al. 2005)。この改変マウスは心臓の形成不
全を示し(Chen et al. 2006),特に心筋繊維の形成
(Dorner et al. 2005)や刺激伝導の異常(Asher et al.
2005Lim et al. 2008)が確認され,CAR が細胞間
接着やコミュニケーションに重要な機能を担っている
と考えられた。また,CAR の siRNA を初代培養のセ
ルトリ細胞に導入すると,接着分子 ZO1 の発現が減
少するとともに膜タンパク質 Occludin がエンドサイ
トーシスにより細胞質に移動してタイトジャンクショ
ンが破壊することが観察された(Su et al. 2012)。一
方,幼仔期と成体マウスを用いた CAR 遺伝子の精巣
特異的なコンデイショナルノックアウトマウスでは,
細胞接着の破壊などの損傷は見られなかった(Sul-

tana et al. 2014)。また,CAR 遺伝子を発現する全て
の組織でコンデイショナルノックアウトを行った成体
マウスでは,正常マウスとの間に顕著な差が見られな
かったが,心臓の房室ブロック,膵臓外分泌腺の委
縮,小腸の拡張および胸腺細胞数の微増などの差が観
察された(Pazirandeh et al. 2011)。
以上のように,遺伝子改変動物や siRNA を用いた
研究から CAR の機能を探る試みがなされているが,
現在までのところ,CAR のノックアウトは胎性致死
を示すという知見以上のものは得られていない。この
ことは,CAR は胎仔期における種々の臓器の発生過
程で重要な機能を担っている事を示している。また,
下垂体の幹細胞ニッチで CAR が発現している事は我
々の報告(Chen et al. 2013)以外にはなく,遺伝子
改変動物における下垂体への影響に関する報告はない。
. CAR の分子レベルでの機能
)細胞間接着
CAR の機能に関する研究は,細胞間接着分子とし
て同定されたことから始まった。まず,CAR 陰性の
株化細胞に CAR を過剰発現させ,CAR 陰性細胞と
共培養する実験が行われた。その結果,CAR 陽性細
胞は CAR 陰性細胞と接着せずに,CAR 分子のホモ
フィリックな接着により形成された細胞集団を形成し
ていることが確認された(Cohen et al. 2001Honda
et al. 2000)。
一方,CAR と相互作用する分子が相次いで同定さ
れ,異種間結合による細胞間接着が確認されている。
ZO1 (Raschperger et al. 2006), Membraneassociat-
ed guanylate kinase 1b (MAGI1b), Protein interact-
ing with protein C kinase (PICK1), PSD95 (Ex-
coŠon et al. 2004),そして,MultiPDZ domain pro-
tein1 (MUPP1) (Coyne et al. 2004)などは CAR と
相互作用する分子として同定され,こうした分子は,
細胞極性の維持とともに,タイトジャンクションやギ
ャップジャンクションなどの細胞間接着装置の形成な
どに関わるものであることが分かった。
CAR が細胞間隙の障壁構造であるタイトジャンク
ションなどの細胞間接着に重要な役割を担っているか
どうかを確かめるために,CAR を強制発現した上皮
細胞を用いて,高分子やイオンの通過率を比較する実
験が行われた(Cohen et al. 2001)。その結果による
と,CAR の発現により,上皮細胞の密着性が強くな
り,蛍光分子 FITC で標識したデキストランの様な高
分子や低分子イオンの透過に対する抵抗性が有意に高
くなることが確認された。CAR によるタイトジャン
クションの形成は,コクサッキーウイルスやアデノウ
イルスへの感染抵抗性を強くする要因となる研究報告
もあり(Cohen et al. 2001),大変興味深い。
)細胞移動
免疫グロブリン様 Ig ドメインを持つ一群のタンパ
ク質は,イムノグロブリンスーパーファミリー
(IgSF)と呼ばれる。このメンバーのタンパク質は,
Ig ドメインを介して細胞間の接着や認識に関わって
いる。
CAR の Ig ドメインである膜外ドメインと結合する
分子として,細胞接着分子の一つである JAML
(Junctional Adhesion MoleculeLike)が同定されて
いる(Zen et al. 2005)。JAML は IgSF のタイプの
膜タンパク質であり,CTX ファミリーに属する分子
である。この JAML は,同種結合以外にも,多数の
類似構造を持つタンパク質と異種間結合を形成する。
最近,血球系細胞における JAML の発現が確認され
(MoogLutz et al. 2003Weber et al. 2007),感染や
外傷の刺激により引き起こされる白血球の炎症性遊走
に機能していることが報告されている(Martin
Padura et al. 1998Zen et al. 2005)。このことから,
CAR が白血球遊走に関与している可能性が推測され,
CAR/JAML の機能解析が行われた(Guo et al. 2009
Zen et al. 2005)。組換え体 CAR をコーティングした
培養用ディッシュを用いて,CAR 陰性/JAML 陽性
の多形核白血球(PMN 細胞)と,CAR 陽性/JAML
陰性の腸管上皮細胞株である T84 細胞と共培養する
実験が行われた。その結果,PMN 細胞が T84 細胞
の形成する細胞層を通過して,経上皮遊走することが
確認された。その際の PMN 細胞の通過は,JAML
や CAR の抗体を添加することにより大幅に抑制され

ることから,CAR と JAML の結合は,タイトジャン
クションが存在する上皮細胞間に白血球を通過移動さ
せるために重要な構造であることが判った(Zen et al.
2005)(図 3C)。その他に,CAR と JAML の結合は
細胞増殖の制御やサイトカイン産生の促進などに関わ
ることが報告されている(Witherden et al. 2010)。
)幹細胞の運命決定と不均等分裂
もう一つの CAR 結合タンパク質として,転写因子
LNX (LigandofNumb proteinX)が同定されてい
る。LNX はユビキチン化能を持っており,幹細胞の
不均等分裂に関与するタンパク質として知られる
Numb に結合する(Dho et al. 1998)。Numb は,発
達する神経系で幹細胞の維持を担っている Notch シ
グナル経路の細胞内伝達を抑制する機能を持ってい
る。そのため,分裂後の片方の娘細胞で Numb が働
くと,前駆細胞への分化が誘導されることになる。さ
らに,Numb は前駆細胞へと誘導された娘細胞の移動
と分裂制御を行うことで(Zhou et al. 2011),その後
の機能細胞への分化を抑制し,前駆細胞群の維持に重
要な役割を担っていることが報告された(Petersen et
al. 2002)。そして,Rice らは,LNX が Numb をユビ
キチン化することにより,タンパク質分解系を誘導
し,細胞内の Numb タンパク質レベルを調整してい
る因子であることを報告している(Rice et al. 2001)。
しかし,幹細胞や前駆細胞におけるこの制御の具体的
な機序に関しては,まだ明らかではない。その後,
CAR が LNX 分子の細胞内領域に存在する第二の
PDZ ドメインと相互作用していることが確認された
(Mirza et al. 2005Sollerbrant et al. 2003)。このこ
とから,CAR は Notch シグナル経路を介する幹細胞
の不均等分裂による娘細胞の運命決定や前駆細胞の補
給と維持に関わる可能性があると考えられる。今後,
その機序の解明が期待される。
)細胞分裂と増殖
複数の研究において,がん組織では,正常組織と比
べて CAR 遺伝子の発現が低下,もしくは消失するこ
とが分かり,CAR 遺伝子の発現レベルの低さががん
の悪性度と関連していることが示されている(An-
ders et al. 2009)。こうした報告から,CAR が,がん
の発生,増殖,転移などの過程に関与している事が推
測され,がんの発生機序の解明や治療法開発に対して,
CAR に関する研究が新たな展開をもたらすことが
期待される。がん細胞株において,CAR はがん細
胞間の接着に関与し(Bruning & Runnebaum 2003),
CAR の発現量に応じて細胞集合が促進される一方
で,がん細胞の増殖が抑制されることが報告されてい
る(Okegawa et al. 2000Okegawa et al. 2001)。こ
の時,CAR の膜外ドメインに対する抗体を加える
と,細胞増殖の抑制効果が有意に抑さえられたが,完
全に抑止することはできなかった。そのため,CAR
の機能は膜外ドメイン以外に,膜内ドメインが関わる
細胞内シグナリングに関与していることが考えられて
いる(Okegawa et al. 2001)。一方,CAR はサイクリ
ン依存性キナーゼ阻害因子 p21 が関与する細胞分裂
サイクルの制御に関わることを示唆する報告がある
(Hall et al. 2000)。CAR の過剰状態では,p21 のタン
パク質レベルが増加すると共に,細胞分裂を抑制する
がん抑制因子 RB(Retinoblastoma)のリン酸化され
た安定型分子の量が増加して,細胞増殖の抑制作用が
亢進された(Okegawa et al. 2001)。いずれの結果も,
CAR が細胞の成長や増殖に負の制御を行っているこ
とを示している。
一方,転写因子 SP1 は GC に富む配列(GC box)
に特異的に結合し,様々な遺伝子の転写を制御してい
ることが知られているが(Li & Davie 2010),CAR
遺伝子の発現が,SP1 によって調節されていること
が報告されている。最近,Chung らは,転写因子
SP1 が CAR のプロモーターを制御していることを示
した(Chung et al. 2011)。確かに,ヒト CAR 遺伝子
のプロモーターの-503/-498領域に,SP1 結合配列
が存在する。ヒストン脱アセチル化酵素阻害剤トリコ
スタチン A(TSA)処理によりクロマチン構造を開
放状態にして SP1 を結合させると,CAR 遺伝子の転
写が活性化された。CAR 陰性細胞株においても,
SP1 の強制発現により CAR 遺伝子の発現増加が確認
された。ところで,SP1 は多くのがん細胞で発現し

ており,腫瘍マーカーの一つとしても考えられてい
る。最近,がん細胞が劣悪な環境に適応するメカニズ
ムの一つとして,SP1 による低酸素応答の活性化が
報告されている(Koizume et al. 2012)。このことは,
CAR と SP1 の関連性を追求することで,がん細胞研
究の新しい糸口になる可能性を示唆している。
. CAR が関わる細胞内シグナル伝達経路
生体内の各々の細胞における機能の多くは,種々の
細胞内シグナル伝達により調節されている。そうした
中に,以下に述べるような CAR と細胞内シグナル伝
達に関する報告がある。
)NFkB 経路と JAKSTAT 経路
腫瘍壊死因子であるTNFa とインターフェロン
IFNg は,動物体内で病原体などに対する感染防
御,免疫,および,炎症の調節に関与するサイトカイ
ンである。TNFa は NFkB (Nuclear Factorkappa
B)の活性化因子であり,抗アポトーシス遺伝子の発
現を抑制して細胞死を誘導する。一方,IFNg はヤ
ヌス・キナーゼ(Janus kinase, JAK)の活性化因子
であり,細胞増殖,分化および生存などの過程を制御
するシグナル伝達兼転写活性化因子 STAT (Signal
Transducers and Activator of Transcription)を調節
している。Vincent らは,内皮細胞や気管支上皮系譜
の細胞株を TNFa と IFNg の存在下で培養すると,
CAR の mRNA とタンパク質のいずれもが著しく減
少することを報告している(Vincent et al. 2004)。こ
のことから,CAR は JAKSTAT 経路を介する細胞
内シグナル伝達系を動員している可能性が考えられる。
)TGFb シグナル経路
TGFb (Transforming Growth Factorb)シグナル
経路に関して,CAR が関与している報告がある。上
皮間葉転移(EpithelialMesenchymal Transition,
EMT)は,上皮細胞から間葉系様細胞へと質的な変
化を起こす現象であり,様々な組織形成とその過程に
おける細胞侵入に関わっている。多くの研究により,
TGFb は Snail, SIP1, Twist, E2A (E47/E12)など
の転写因子を動員して,上皮接着分子の転写を抑制し,
EMT を介して細胞の遊走を誘導する因子であること
が示されている(Peinado et al. 2004Wang et al.
2013)。そのほか,細胞増殖や分化にも機能している
こと(Ten Dijke et al. 2002)や,上皮系細胞には強
い増殖抑制作用(Sancho et al. 2004)を示すほか,
他の細胞種には促進作用も見られた(Civinini et al.
2013)。
EMT の過程において,上皮細胞におけるタイトジ
ャンクションの機能を担っている CAR と Ecadherin
の挙動を調べた実験がある。TGFb の下流にある転
写因子 Snail が,CAR と Ecadherin の遺伝子上流の
プロモーターに直接結合することにより,転写抑制を
行い,EMT を誘導することが確認されている。
TGFb により EMT の誘導を行うと,CAR と E
cadherin の 遺 伝 子 発 現 は い ず れ も 抑 制 さ れ た
(Lacher et al. 2006)。
TGFb シグナル経路を介した CAR 遺伝子の転写
抑制の制御には,転写因子 ZEB1 との協同作用が必
要である。ヒト膵臓細胞株 PANC1 と乳腺細胞株
MDAMB231 を用いて ZEB1 遺伝子の発現をノッ
クダウンすると,TGFb に誘導される CAR 遺伝子
の転写抑制効果が抑えられた(Lacher et al. 2011)。
この抑制は,CAR 遺伝子の転写開始点上流に存在す
る E2 box への ZEB1 の結合を介する直接の制御であ
り,ZEB1 遺伝子の過剰発現による CAR 遺伝子の発
現低下の確認実験からも証明されている。ところが,
TGFb の誘導による ZEB1 タンパク質の量の変動は
起こっていなかったため,ZEB1 は TGFb シグナル
経路と協同的に CAR 遺伝子の発現制御に機能してい
ると予想されている(Lacher et al. 2011)。
以上の様に,CAR が EMT に関与することは判っ
たが,CAR の減少と細胞運動性の獲得に関与する直
接的な証拠は未だにない。細胞増殖や分化に関与して
いる可能性も含めて,今後の研究が期待されるところ
である。
)Ras/Raf/MEK/ERK シグナル経路

によって制御されているとの報告がある(Anders et
al. 2003)。Ras/Raf/MEK/ERK は,細胞の増殖・分
化・アポトーシスにおいて中心的な役割を担っている
シグナル伝達系である。多くの転写因子を介して,細
胞増殖,分化の促進やアポトーシスの抑制などを引き
起こす。さらに,このシグナル経路は,過剰活性化な
どの異常による細胞の癌化と関連し,その過程におけ
る接着分子の遺伝子発現に対する負の制御も示唆され
ている(Chen et al. 2000)。MAP キナーゼキナーゼ
(MEK)のインヒビター U0126 と PD184352 を結腸
直腸癌細胞株 SW480 に添加すると,この細胞におけ
る CAR 遺伝子の発現は有意に増加した(Osabe et al.
2009)。これに対して,マメ科ネムノキ亜科の若葉や
芽に含有されている毒性アミノ酸ミモシンで処理し
て,細胞サイクルを G1 に停止させて細胞分裂を止め
ると,CAR 遺伝子の発現レベルは変化しなかった
(Anders et al. 2003)。一方,抑制因子 PI3K を増加さ
せて Ras の働きを抑えると,CAR 遺伝子の発現が促
進された。逆に,Raf1 を活性化すると,CAR 遺伝
子の発現の減少が確認された(Anders et al. 2003)。
以上のことから,CAR 遺伝子の発現が Ras/Raf/
MEK/ERK 経路の活性化によって負の制御を受けて
おり,CAR が細胞接着,増殖,分化などの過程に関
与していることが示唆される。しかし,CAR 遺伝子
の発現を直接制御する因子や分子機序は不明であり,
今後の研究が待たれる。
. あとがき
これまでの研究で,CAR は細胞膜受容体として,
細胞間の相互作用に応じて働く様々なシグナル伝達系
を介し,器官形成や細胞成長,増殖などの重要な生理
機能に関与していることが明らかになってきた。特
に,個体発生において,CAR 遺伝子は,胎仔期の未
成熟の組織原基で高発現していること,成熟に伴い発
現が減少していること,さらに,成体の神経幹細胞ニ
ッチに CAR 陽性細胞が存在することが確認された
(Hotta et al. 2003)。著者らは,最近,下垂体の幹細
胞ニッチと考えられている Marginal Cell Layer(前
葉と中葉が面する互いの一層の細胞群)と,前葉の実
質層に CAR 陽性細胞が存在することを報告した
(Chen et al. 2013)。CAR が幹細胞ニッチ形成に関わ
っているとの報告は,組織の維持の視点で見ると大変
興味深いものである。その機能を分子レベルで解明す
ることは,胎仔期の組織原基の形成における役割と共
に,成体の組織形成,そして,がんの治療や移植組織
の再生を含む再生医療などの重要な課題に答えること
になると期待される。本稿を踏まえて,我々が下垂体
とそれ以外の組織で観察した CAR 陽性細胞について
は,稿を改めて述べたい。
