アストログリアに関する研究では、これまでに次のことを
明らかにした。①アストロサイトにおいて酸化ストレス緩和
に関与する遺伝子群(oxidoreductase genes)の発現をATPが上昇させた。②ATPは、酸化ストレス過酸化水素誘発アストロサイト細胞死をP2Y1 受容体を介して抑制した。③過酸化水素誘発アストロサイト細胞死の詳細な機序を明ら
かにした。④金属イオン依存性蛋白質分解酵素-9(MMP-9)は細胞外マトリックスを除去し、細胞の増殖、突起進展の際
に空間を提供する非常に重要な分子であるが、一方で種々の
細胞外分子を分解するために、BBB 破壊等の細胞外環境破壊も引き起こす。ATPはアストロサイトのTNF-alpha産生能をtonicに抑制することにより、MMP-9産生量を調節し、脳内恒常性の維持に関与しているものと考えられた。⑤glio-transmitterであるATPがP2Y1受容体活性化を介したアストログリアの細胞死回避・機能維持により、実際に酸化ストレ
ス誘発性の神経細胞死を防御することを明らかにした。さら
に、アストログリア細胞のP2Y1受容体活性化によりサイトカイン IL-6の放出が促進され、ATP刺激アストログリアの神
経保護効果が IL-6中和抗体共添加により抑制されたこと,さらに外因的に加えた IL-6がH2O2誘発障害に対して神経保護作用を示すことから、ATPはアストログリアからの IL-6産生・放出機能を促進することにより神経細胞を保護すること
を明らかにした。このように、これまでの脳保護薬とは異な
ったアストログリアのATP受容体をターゲットとした全く新規の創薬の一助となる可能性がある。
ミクログリアに関しては次の成果がある。痛みを伝える末
梢神経の損傷や機能異常は神経因性疼痛という耐え難い慢性
疼痛を引き起こす。しかし、その発症維持機構は依然不明で
あり、モルヒネなどの鎮痛薬が奏功しがたく、有効な治療法
もない。我々は、末梢神経の損傷後に脊髄で活性化したミク
ログリアにおいてイオンチャネル型ATP受容体サブタイプP2X4受容体が過剰発現し,その受容体刺激が神経因性疼痛に重要であることを示した(Nature, 2003)。さらに、本プロジェクトにおいて、P2X4受容体の活性化によりミクログリアからBDNFが放出され、TrkB受容体を介する陰イオン勾配の変化によってGABAニューロンが抑制性から興奮性へと

図 解き明かされた神経因性疼痛の発症メカニズム末梢神経の損傷後に脊髄で活性化したミクログリアにおいてイオンチャネル型ATP受容体サブタイプP2X4受容体が過剰発現し,その受容体が刺激される。さらに、P2X4受容体の活性化によりミクログリアからBDNFが放出され、TrkB受容体を介する陰イオン勾配の変化によってGABAニューロンが抑制性から興奮性へと変化し、疼痛に関連する神経興奮を引き起こす。

(Nature, 2005)。以上の成果は、P2X4受容体から端を発するミクログリア-ニューロン連関の重要性を示唆してお
り、神経因性疼痛の新しいメカニズムとして注目されてい
る。その後更に研究を進め、このメカニズムに重要なP2X4受容体発現増加機序とミクログリア活性化機構についても明
らかになりつつある。また、我々は、細胞外マトリックス分
子であるフィブロネクチン(FN)がP2X4受容体発現を増加することを見出した。FNはミクログリアに発現するインテグリンを介してSrcファミリーキナーゼ(SFK)を活性化させ、P2X4受容体発現レベルを増加させた。FNは神経因性疼痛ラットの脊髄後角で増加し、インテグリン拮抗薬により
P2X4受容体の増加とアロディニアが共に抑制された。したがって,FN/インテグリンシグナルがミクログリアでのP2X4受容体発現増加機構の一端を担っていることが示唆された。
ミクログリアのもう一つの大切な役割である「脳内のお掃
除屋さん」については不明の部分が多く、ミクログリアがど
のように活性化されて貪食作用を発揮するのかはよく分かっ
ていなかった。本プロジェクトにより、ミクログリアにATP受容体サブタイプの一つP2Y6受容体が発現していることを免疫組織学的、薬理学的手法によりあきらかにし、次いで、
そのP2Y6受容体を選択的アゴニストUDPで刺激すると、細胞内カルシウムの上昇と共に、ミクログリアの貪食能が飛躍
的に増強することを証明した(in vitro)。また、丸ごとの動物を使った実験(in vivo)では、けいれん誘発物質のカイニン酸をラットの腹腔内に投与して、海馬領域での神経細胞
死を確認し、その標本では、ミクログリアが高度に活性化さ
れ、またP2Y6受容体の発現が非常に高まっていることを明らかにした。ミクログリアの刺激を引き起こすUDPが、カイニン酸により放出されることを、in vitroでも in vivoでも証明することができた(Nature 446, 1091-1095, 2007)。
