Research
The year 2019 marks my eighth year of study in the lab. I’ve
researched ichthyosis under Dr. Nomura. I’ve recently been
focusing on investigations into patients with unexplained ich-
thyoses that cause revertant mosaicism and into patients with
Nagashima-type palmoplantar keratosis whose pathogenic
mutation hasn’t been identified. Although I’ve used every
method in molecular biology from traditional to cutting-edge,
I haven’t got the answer. I’d like to elucidate the mechanism
definitively this year.
2019年で8年目になります。乃村先生のもと、魚鱗癬の研究を推進しております。最近は
「revertant mosaicismを伴う原因不明の魚鱗癬患者」と「長島型掌蹠角化症の中で、病原遺伝子変異が特定できていない患者」の解析に力を入れております。これまで古典的なものから最新鋭のものまで、分子生物学のあらゆる手法を駆使しておりますが、正解に辿り着けておりません。今年こそはなんとかしたいと思っております!

私のチームでは、引き続き、魚鱗癬や掌蹠角化
症を切り口に、遺伝性疾患全体の発展に繋がるよ
うな研究を志向しています。具体的には、リード
スルー治療や変異mRNA分解機構、体細胞にお
ける組換え誘発機構などを研究しています。最
近は良い論文が出始め、2016年はJID、2018
年はJCI InsightとJID、この原稿を書いている
2019年1月にはLife Science Allianceに研究
成果が掲載されました。2019年はとっておき
のネタ(?)がありますので、さらに良い論
文が出るものと確信しています。遺伝性疾患
の診断や治療を前進させるような新しい科学
的知見を得ることを目標に、チームのメン
バー全員で力を合わせて頑張りたいと思いま
す。そして、患者さんの会(魚鱗癬の会ひま
わり)との連携もさらに密にし、患者さんに
今すぐ直接的に役立つような活動をチームで
行いたいと思います。
My team conducts research with the aim of developing treat-
ments for the general hereditary disorders by using ichthyosis
and keratoderma as a model disease. More specifically, our
research focuses on read-through therapy, mRNA decay and
somatic recombination. In the past few years, some of our
results have been published in the Journal of Investigative
Dermatology (JID) in 2016, in JCI Insight and the JID in 2018,
and in Life Science Alliance in January 2019. We are now pre-
paring a new paper which will be submitted to even better
journals. Our ultimate goal is
to gain new scientific knowl-
edge that will be helpful for
diagnosing and treating
hereditary disorders. Also,
we’d like to contribute more
and more to the Japanese
patient organization for ich-
thyosis called “Himawari”.

By the time the 2018 annual report is published, I hope to have
earned my master’s degree. I’ve been working on experiments
relating to readthrough treatments, which I hadn’t done for
a long time, and I’m considering the results of whole exome
sequencing. I feel strongly that I can never study too much. The
world is surrounded with things I don’t know. I’ll try keeping at
my own pace, even though I’m busy with many things.
In April 2018, I opened a whole new chapter of my life
by joining Team Nomura as a first-year doctoral student.
Under the guidance of the whole team, I’ve been actively work-
ing on compound screening for Nagashima-type palmoplantar
keratosis. This disease is known to result from a nonsense muta-
tion, and readthrough therapy has been proposed as a potential
cure. By screening compounds, I aim to identify the potent
ones which are able to readthrough the mutation. Being new
It’s been four years since I began my research on the mecha-
nism of revertant mosaicism. This promising line of research
has the potential to lead to treatments for hereditary skin
disorders and to elucidate the mechanism behind the appear-
ance of skin tumors. I’ve gotten positive results through
various analyses. The chance to meet patients at the Special
Clinic for Ichthyosis and Keratoderma motivates me in my
research. I’d like to apply this experience to that research.

最近は久しぶりのリードスルー関連実験、Whole exome sequencingの結果の検討などをしています。ここ数年、勉強はしてもしてもキリがないなぁと強く感じております。世の中わからないことだらけです。今年も相変わらずバタバタしそうですが、自分のペースを崩さずにうまくやっていければなぁと思っております。

planning. It’s been almost a year
since I started doing research, and
I’m glad to be constantly learn-
ing and gaining knowledge every
single day. Life in Japan has been
fulfilling: I’m always occupied with
the research work that I’m interested in. I’m truly
grateful for all the opportunities given to me by the
university and the department. The seasons and
everything else I’ve experienced so far have been
lovely. I’m definitely looking forward to what lies
ahead in the coming years.
Revertant mosaicismの機序解明を目指して研究を開始し、4年が経ちました。遺伝性皮膚疾患の治療や皮膚腫瘍出現のメカニズムにも繋がり得る夢のある研究テーマで、色々な解析を通じて少しずつポジティブな結果も得られてきました。魚鱗癬掌蹠角化症外来で実際に患者さんに接することは研究のモチベーションにも繋がりますし、今後もそこで得られる経験に基づいた研究を続けていきたいと思います。
